Research Context
MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino-acid mitochondrial-derived peptide (MDP) encoded within the mitochondrial 12S ribosomal RNA gene, first described in 2015. Unlike the vast majority of human peptides, which are encoded by nuclear DNA, MOTS-c is one of a small class of MDPs encoded directly by the mitochondrial genome. The class also includes the humanin family and the small humanin-like peptides (SHLP1 to SHLP6). MOTS-c is detectable in plasma, declines with age, and has become the prototypical mitokine in retrograde mitochondrial-to-nuclear signalling research.
Preclinical literature has examined MOTS-c as a tool compound for investigating mitochondrial metabolism, AMPK-mediated metabolic flexibility, insulin sensitivity in skeletal muscle and adipose tissue models, and exercise-mimetic signalling pathways. It is widely cited in studies of cellular energy homeostasis and ageing biology.
