Melanotan-1 (also designated [Nle4, D-Phe7]-α-MSH or NDP-α-MSH) is a synthetic tridecapeptide analogue of the endogenous hormone α-melanocyte-stimulating hormone (α-MSH). Two substitutions distinguish it from native α-MSH: norleucine (Nle) at position 4 and D-phenylalanine at position 7. Together, these substitutions confer resistance to enzymatic degradation and elevated binding affinity at the melanocortin-1 receptor (MC1R) relative to the parent hormone in cell-based assay systems.

The N-terminus is acetylated and the C-terminus is amidated, modifications standard for melanocortin agonist analogues that block exopeptidase degradation. Lyophilisation & Stability
