Research Context

KPV is the synthetic tripeptide L-lysyl-L-prolyl-L-valine (Lys-Pro-Val), corresponding to residues 11 to 13 of alpha-melanocyte-stimulating hormone (alpha-MSH). The full alpha-MSH peptide is a 13-residue post-translational cleavage product of pro-opiomelanocortin (POMC) and acts through the melanocortin receptor family. Carboxy-terminal tripeptide fragments of alpha-MSH, including KPV, were characterised in early melanocortin structure-activity studies as retaining a subset of the parent hormone’s biological signatures despite lacking the melanocortin receptor-binding pharmacophore at the N-terminus.

Preclinical studies have examined KPV in models of mucosal inflammation, intestinal-epithelial signalling, NF-kappaB pathway modulation, and cellular responses associated with the alpha-MSH C-terminal motif. Patents covering antipyretic and anti-inflammatory compositions of Lys-Pro-Val were filed in the early 1990s, and the tripeptide has remained an active reference compound in melanocortin and inflammation research.

Chemistry & Structural Modification

KPV is an unmodified linear tripeptide of three standard L-amino acids, joined by conventional alpha-peptide bonds, with free N-terminal amine and C-terminal carboxylic acid (H-Lys-Pro-Val-OH). No protecting groups, lipidation, or cyclisation.

Lyophilisation & Stability

KPV is supplied as a lyophilised white powder, sealed under controlled conditions. Lyophilisation removes residual moisture that would otherwise promote slow peptide-bond hydrolysis during long-term storage. As a short, unmodified linear tripeptide with no cysteine, methionine, or tryptophan, KPV has no obvious oxidation-prone residues, but standard cold-and-dry storage remains best practice. Sealed vials kept cold and dry retain ≥99% potency well within the labelled shelf life.
