Research Context

SLU-PP-332 is a small-molecule research compound, not a peptide. It is a synthetic aromatic hydrazide acting as a pan-agonist of the estrogen-related receptor (ERR) subfamily of nuclear receptors. The ERRs (ERRα, ERRβ, ERRγ) are orphan nuclear receptors structurally related to the classical estrogen receptors but with distinct ligand-binding pockets and constitutive transcriptional activity; they regulate metabolic gene programmes including mitochondrial biogenesis, oxidative phosphorylation, and fatty-acid oxidation. SLU-PP-332 features a naphthalene-substituted aromatic scaffold that yields approximately 50-fold improved ERRα potency relative to earlier-generation ERR ligands.

Preclinical studies have examined SLU-PP-332 as a chemical tool for investigating ERR-driven transcriptional programmes, mitochondrial function and cellular respiration in cultured cell models.

Chemistry

SLU-PP-332 is a small-molecule aromatic hydrazide bearing a 4-hydroxybenzene ring connected through a hydrazide linkage to a 2-substituted naphthalene moiety. Formal name: (E)-4-Hydroxy-N’-(naphthalen-2-ylmethylene)benzohydrazide. The planar aromatic scaffold is designed to make pi-pi stacking contacts within the ERR ligand-binding domain.

  • Molecular formula: C18H14N2O2
  • Molecular weight: 290.32 g/mol
  • CAS number: 303760-60-3; PubChem CID 5338394
  • Reported in-vitro activity: ERRα EC50 ~98 nM, ERRβ ~230 nM, ERRγ ~430 nM (cell-based cotransfection reporter assays).

Lyophilisation & Stability

SLU-PP-332 is supplied as a lyophilised white to off-white powder, sealed under controlled conditions. Although it is a small molecule rather than a peptide, lyophilisation and sealed-vial packaging still remove residual moisture and oxygen exposure that would otherwise drive slow degradation of the hydrazide linkage during storage. The hydrazide chemistry is sensitive to prolonged exposure to strong acid or strong base; the supplied lyophilised solid is stable for the labelled shelf life under cold, dry, sealed conditions.

Storage

Store lyophilised vial at 2 to 8°C, protected from light, in the original sealed vial. (Long-term storage at −20°C is standard practice.)

Reconstituted/dissolved solution: 2 to 8°C if used short term. The originating literature notes that DMSO stock solutions of SLU-PP-332 may be stored at −20°C for up to 3 months. Avoid repeated freeze–thaw cycles.
